ISBt1 putative transposase is 33 % identical (over a 250 residues overlap) to those of IS1221A from Mycoplasma hyorhinis GDL-1, IS861 from Streptococcus agalactiae COH-I, and IS1296 from Mycoplasma mycoides subsp. mycoides L2. Interestingly, ISBt1 is apparently lacking the "expected ORF1" observed in the other IS3 family members. This is reflected in its small size. ISBt1 is located downstream of the CryID gene and 92 bp upstream of the ATG of the CryIC toxin gene, as a single copy in the various plasmids studied. Note that in Bacillus thuringiensis subsp. fukuokaensis 84I113 (Dunn and Ellar, 1997; Accession Number Y09946), two overlapping ORFs (OrfX and OrfY) display similarities to other IS3 family member transposases, and OrfY is most similar to the ORF of ISBt1. However, no IR were found flanking these ORFs.
