IS493 transposase is 36 % identical to that of IS1373. Although two ORF are present within IS493, 177 (179-712) and 254 (864-1628), the second one has been shown to be the actual transposase (Baltz et al., 1992). This IS might just be a complex (rearranged) element. Several vectors have been derived from IS493: Tn5096 to Tn5099 (xylE promoter probe) and Tn5099-10, a derivative that transposes at a 1000-fold higher frequency, due to a change in its left IR (Solenberg and Baltz, 1994). The target consensus results from the analysis of about 15 different sites.
