IS4 was originally detected at a single site only within the gal operon (quoted in Habermann et al., 1979). There is only one copy of IS4 in Escherichia coli BHB2600 (mapped by Birkenbihl and Vielmetter, 1989), HB101, and W3110. Within the ECOR collection, the IS4 copy number varies from zero to 14, with an average of 5 copies/strain (Dykhuizen et al., 1985). An IS distribution study among 23 closely related O1:K1 pathogenic E. coli strains showed an average of 9 IS2 copies/genome, as compared to the 41, 17, 4, and 3 copies/genome for IS1, IS4, IS5 and IS30, respectively (Lawrence et al., 1989). Using hybridization, IS4 was not detected in strains of Salmonella (6 strains), Citrobacter freundii (1), Enterobacter aerogenes (1), Klebsiella aerogenes (1), Klebsiella pneumoniae (1), Proteus vulgaris (1), Proteus mirabilis (1), Serratia marcescens (1), but in Shigella sp. ATCC11126 (Lam and Roth, 1983). In Escherichia coli minicells, a 47,000 protein corresponding to IS4 transposase has been observed in small amounts (Trinks et al., 1981).
