IS256 is associated with the aacA-aphD gene coding for a bifunctional enzyme specifying resistance to gentamycin, tobramycin and kanamycin, in a composite transposon structure. Its name is Tn5281 in the conjugative plasmid pBEM10 from Enterococcus faecalis HH22 (Hodel-Christian and Murray, 1991), Tn4001 in the Staphylococcus aureus plasmid pSK1 (Lyon et al., 1984), and Tn4031 in Staphylococcus epidermidis (Thomas and Archer, 1989). Multiple copies of IS256, not associated with any resistance genes, are present in the chromosome of Staphylococcus aureus BM3121 (Dyke et al., 1992), but also in many strains of Enterococcus faecalis and E. faecium (Rice and Thorisdottir, 1994). In Enterococcus faecalis CH19, the chromosomal beta-lactamase gene is located ca. 600 bp downstream of an IS256 copy (IS256E), and upstream of an iso-ISS1W (Rice and Marshall, 1994). The IS256-based transposon Tn4001 has been used in a suicide vector (pISM1001) for random insertion in Mycoplasma gallisepticum (avian mycoplasmosis), with efficiency of 10-6 transposant/CFU (Cao et al., 1994), but also in Acholeplasma oculi ISM1499 (Knudtson and Minion, 1993) and Streptococcus gordonii (Lunsford, 1995). Vancomycin type B (VanB) is mediated by the vanR, S, Y, W, B, X gene cluster which directs synthesis of peptidoglycans precursors terminating in D-alanyl-D-lactate. This cluster is located within the 64-kb composite transposon Tn1547, composed of IS16 and an IS256-like (2 nt different than the original IS256 from Tn4001 of Staphylococcus aureus) element in a direct orientation (Quintiliani and Courvalin, 1996).
