ISH26 has been isolated through its inactivation of the bacteriorhodopsin (BR, bop) gene. There are 4 ISH26 copies on pHH1, and 4 copies on the chromosome of Halobacterium salinarum PHH1 (Ebert et al., 1986). ISH26 was described as harbouring two overlapping ORFs. Although the first ORF shares significant similarity with the putative transposases of other IS4 family members (26 % identity with IS231W, over a 143-aa overlap), the second ORF only shares very limited similarity, in the region of the conserved "E" residue. Detailed analyses suggest however that several frameshiftings could significantly increase this similarity. The first open reading frame is very closely related to the N terminal end of the Tpase of the different ISH8. Therefore, a reevaluation of ISH26 DNA sequence is needed to clarify this issue.
