ISPsp10 is 86% aa similar to ISPst8. The third ORF is a putative ORFAB transposase reconstructed in silico by a possible -1 frameshift. ISPsp10 has been identified by its transposition to entrapment vector pMAT1 (sacB cassette). A unique feature of ISPsp10 is the lack of a stop codon within ORFB encoding the terminal part of the transposase. ISPsp10
transposed into the sacB gene just upstream of a +TAA sequence (upon insertion the dinucleotide TA was duplicated); therefore, the stop codon in this case is provided by the target site of transposition. As judged from DNA hybridization analysis ISPsp10 is present in 4 copies in the genome of Pseudomonas sp. LM25. Homologous sequences were also detected by DNA hybridization in Pseudomonas sp. LM15.
