ISBth5 was found by screening completely sequenced genomes for sequences homologous to the IS4 transposase using BLASTP. Multiple sequence alignments revealed a conserved DDE motif : D(N2)-70-D(N3)-150-E(C1). ISBth5 is located on plasmid pBT9727. It's copy number in Bacillus thuringiensis serovar konkukian str. 97-27 is 1. In addition to the transposase ORF, ISBth5 harbours three passenger genes that are almost identical to those of IS231L (a probable metalloprotease (ORF1), an uncharacterised conserved protein (ORF2) and a potential transcriptional regulator (ORF3), the latter displaying similarities with TetR, the inducible repressor that regulates expression of tetracycline resistance determinants).
